Ruxolitinib is an orally bioavailable, potent and selective inhibitor of Janus-associated kinase (JAK) 1 and 2, with IC50 to be 2.7, 4.5 and 332 nM for JAK1, JAK2 and JAK3 respectively. Ruxolitinib acts by blocking the JAK/STAT pathway. Ruxolitinib is a drug for the treatment of intermediate or high-risk myelofibrosis, a type of bone marrow cancer. It is also being investigated for the treatment of other types of cancer (such as lymphomas and pancreatic cancer), for polycythemia vera, and for plaque psoriasis. Ruxolitinib has entered in a Phase II clinical trial for the treatment of breast cancer.
